Предварительную визуальную диагностику вируса Шарки сливы проводили в насаждениях косточковых культур (слива, алыча, вишня, черешня) в период вегетации: в конце весны – начале лета. Оценена результативность диагностики белорусских изолятов вируса Шарки сливы с помощью ряда лабораторных методов, в том числе с использованием поликлональных, рекомбинантных и моноклональных универсальных антител для иммуноферментного анализа и метода IC-RT-PCR.</p><p>Показано, что поликлональные антитела более эффективны для массовой диагностики белорусских изолятов вируса Шарки, чем моноклональные универсальные антитела.</p><p>В то же время вариант ИФА с использованием поликлональных антител наименее чувствительный, максимальное разведение, при котором было возможно определить наличие данного вируса в тканях растения, не превышает 1:20. Использование моноклональных и рекомбинантных антител в ИФА-тесте позволило определить вирус в образцах, разведенных до 1:100, метод IC-RT-PCR – при разведении экстрактов до 1:50000.</p></abstract><trans-abstract xml:lang="en"><p>The investigations were made within 2011-2012 in the department of biotechnology of the Institute for Fruit Growing. Preliminary visual diagnosis of plum pox virus made at the plantations of stone fruit crops such as plum, cherry plum, sour cherry and sweet cherry ones during their vegetation, i.e. in the end of spring and in the beginning of summer. Diagnosis efficiency of Belarusian isolates of plum pox virus by means of a number of laboratory methods, including use of polyclonal, recombinant and monoclonal universal antibodies for enzyme immunoassay and IC-RT-PCR method was estimated.</p><p>It is shown, that polyclonal antibodies are more effective for mass diagnosis of Belarusian isolates of plum pox virus, than monoclonal universal antibodies.</p><p>At the same time the variant of enzyme immunoassay with use of polyclonal antibodies is the least sensitive. The maximum dilution, at which it has been possible to define the presence of the given virus in plant tissues, does not exceed 1:20.
